What *do* a tick and the Eiffel Tower have in common? is correct. It is a compound subject, therefore it will take a plural verb. You can test this by asking "what do *they* have in common?"

@JohnWain Haha! in order for #2 to be right, you would have to change it to say, "what does a tick have in common *with* the Eiffel Tower?" That way the tick is a single subject that would take the single verb. ("what does *it* have in common...")
