There are a few differences:
* Spelling (e.g 'ized' vs. 'Ised' endings, such as 'paralysed/paralyzed. UK uses S here. America uses Z. Other spelling differences exist).
* Pronunciation is different between the two (although both the UK and US have various accents). Both are mutually intelligible.
* A few words are different. For example, 'rubbish bin' (UK) vs 'trash can' (US).
* Some (though not very many) words have different meanings. For example, 'pants' in the UK means underwear. In the US, 'pants' mean trousers.
That's all I can think of for now, I hope it helps! :)

There's too many differences. I suggest you look on google for more examples but here are some I just thought of off the top of my head:
The use of "vacation" and "holiday"
"Pissed" in America means mad and in the U.K. It means drunk

Pronunciation due to accents is a big difference, but there's a lot of slight spelling differences too here are some examples: (uk will be 1st us 2nd)
Mum - mom
Colour - color
Favourite - favorite
Vapour - vapor
But if you spelt or pronounced a word the USA way in UK or AUS we would all understand it :)
